These peptides were found not to demonstrate direct antioxidant activity but be able to restrict lipid peroxidation of human lipoproteins by modification of their structure. The short peptides increase stability of red blood cell membranes toward osmotic hemolysis. They also elevate the stationary level of intracellular reactive oxygen species and at the same time decrease (all excepting epitalon) percent of dead cells in neuronal population. The suggestion was made that under in vivo conditions, short peptides may participate in apoptosis/necrosis regulation.

The effect of synthetic peptide bioregulators (Epitalon, Livagen and Vilon) on structural and facultative heterochromatin of cultivated lymphocytes have been studied among old (75-88yr.) people. The data obtained indicate that epitalon, livagen and vilon: 1) activate synthetic processes, caused by reactivation of ribosomal genes as a result of deheterochromatinization (decondensation) of nucleolus organizer regions; 2) induce unrolling (deheterochromatinization) of total heterochromatin; 3) release genes repressed by heterochromatinization (condensation) of euchromatic regions forming facultative heterochromatin; 4) epitalon and livagen induce deheterochromatinization (decondensation) of pericentromeric structural heterochromatin of the chromosomes1 and 9. However, vilon does not induce deheterochromatinization of pericentromeric structural heterochromatin. These results indicate that peptide bioregulators Epitalon, Livagen and Vilon cause activation (deheterochromatinization) of chromatin in lymphocytes of old individuals.

Pinealon (Glu-Asp-Arg) has the most pronounced effect among them. The capability of pinealon to increase the neuronal resistance to hypoxic stress in experiments with prenatal hypoxia has a complex nature. It is based not so much on the inhibition of ROS increase in cells in response to stress as on stimulation of internal antioxidative enzyme system and possibly limiting the excitotoxic effect of N-methyl-D-aspartate.

The application of Epitalon in Campbell rats is found to intensify the bioelectric and functional activity of the retina due to the preservation of its morphological structure. Epitalon therapy in patients with degenerative retinal lesions results in a positive clinical effect in 90% of the cases. The analysis of Epitalon effects suggests that the tetrapeptide participates in the mechanisms of transcription common for the epiphysis and retina.

Experimental data on the effect of peptides--Vilon (Lys-Glu) and Epitalon (Ala-Glu-Asp-Gly)--on induction of urinary bladder tumors in rats are presented. Treatment with Vilon was followed by a significant fall in tumor incidence in 56% of experimental animals, as compared with 75.5% in control, as well as inhibition of early-onset neoplastic changes in the bladder mucosa. No inhibitory effect of Epitalon was recorded.

The molecule lacks aromatic residues, so it absorbs weakly near 280 nanometres, the wavelength applied to many other peptides. The peptide bond absorbs strongly below 220 nanometres, making 214 nanometres a practical compromise. Gradient methods must therefore use mobile phases with low ultraviolet absorbance to keep the baseline clean.
Neutral and slightly basic aqueous conditions promote aspartimide formation at the aspartate-glycine step, often followed by deamidation. Elevated temperature and repeated freeze-thaw cycles accelerate the loss. Acidic solutions and dry lyophilised powder are comparatively stable over short periods.
No monograph exists in the United States Pharmacopeia or the European Pharmacopoeia, and no widely distributed certified reference standard is available. Purity and identity therefore rest on supplier methods that are not harmonised. This absence is the main reason reported purities are difficult to compare between sources.
Mass spectrometry provides the identity check, because the observed mass is compared with the theoretical mass of the AEDG sequence. Chromatography separates and quantifies impurities but does not by itself prove which peptide is present. The two techniques are normally used together.
