No pharmacopoeial monograph exists for this peptide, so quality rests on the supplier's internal specifications and on whatever independent testing a purchaser arranges. Certificates of analysis differ widely in which tests they report and in the limits applied. The counter-ion introduced during purification, commonly acetate or trifluoroacetate, changes the net peptide content of a given mass of powder, so two samples of equal weight may not contain equal amounts of the active sequence. Third-party laboratories can verify purity, identity, residual solvents, and counter-ion content for a fee, which makes documentation more informative than labelling.

Many of these papers involve small groups, lack blinding or placebo comparison, and are difficult to retrieve through indexed databases. Review articles published in English generally summarise the claims without reanalysing the underlying data. Because no large randomised trial exists, the clinical importance of these reported effects stays unresolved and is properly described as an open question.
No national medicines regulator has approved epitalon as a therapeutic product. It is generally distributed as a research chemical, and in some jurisdictions selling peptides for human consumption without approval is restricted or prohibited. Certificates of analysis accompanying commercial material vary in which tests are performed, and independent verification of identity and purity is uncommon. Statements about anti-ageing or disease-prevention benefits on vendor pages are marketing claims rather than regulatory findings, a distinction that shapes how the compound is discussed in scientific and popular sources alike.
The most frequently cited laboratory finding is that AEDG increased telomerase activity and extended telomere length in cultured human somatic cells. That work used fetal fibroblast strains and reported changes in enzyme activity alongside altered division counts. Replication by unrelated groups has been limited, and the published record is largely a single-laboratory series rather than a multi-centre programme. The result supports a hypothesis about peptide influence on gene expression in cell culture; it does not by itself establish an effect on telomere length in living animals or in people.

Ageing, melatonin, epithalon (tetrapeptide Ala-Glu-Asp-Gly) and different light conditions effects on protein content and cathepsins B and D activities in rat liver and kidneys lysosomal fractions were studied. Ageing leads to decrease of cathepsins activity in rat liver lysosomal fractions. Constant light and darkness conditions result in earlier age decline of cathepsins activity. Absence of day and night succession in comparison with alternating light conditions causes decline of both general and specific cathepsin D activity. Melatonin and epithalon administration resulted in decrease of cathepsin D activity in liver only under control interchangeable light conditions. Cathepsin B activity in liver and kidneys lysosomal fractions declined in all experimental light conditions. Cathepsins activity decrease under the influence of epiphysial factors is evidently connected with their inhibitory effect on protein and general metabolism.

In investigations carried out with organotypic culture of mediobasal preoptic area (MPA) of hypothalamus it was established that pineal peptide preparations epitalon (2 ng/ml) and epithalamin (100 ng/ml) stimulate the development of proliferative activity of explants in 3 month and 24 months female rats. It has been shown that epithalamin is more effective in young rats in comparison with old animals, but epitalon as well as epithalamin have almost the same less pronounced inducing effect on growth zone in MPA explants from young and old animals. This effect is tissue specific and could be dependent on inhibition of apoptosis.

The effect of synthetic peptide bioregulators (Epitalon, Livagen and Vilon) on structural and facultative heterochromatin of cultivated lymphocytes have been studied among old (75-88yr.) people. The data obtained indicate that epitalon, livagen and vilon: 1) activate synthetic processes, caused by reactivation of ribosomal genes as a result of deheterochromatinization (decondensation) of nucleolus organizer regions; 2) induce unrolling (deheterochromatinization) of total heterochromatin; 3) release genes repressed by heterochromatinization (condensation) of euchromatic regions forming facultative heterochromatin; 4) epitalon and livagen induce deheterochromatinization (decondensation) of pericentromeric structural heterochromatin of the chromosomes1 and 9. However, vilon does not induce deheterochromatinization of pericentromeric structural heterochromatin. These results indicate that peptide bioregulators Epitalon, Livagen and Vilon cause activation (deheterochromatinization) of chromatin in lymphocytes of old individuals.

We have studied the effect of synthetic peptide Epitalon on the activity of ribosomal genes, denaturation parameters of total heterochromatin, polymorphism of structural C-heterochromatin and the variability of facultative heterochromatin in cultured lymphocytes of persons aged 76-80 years. RESULTS: The obtained data demonstrate that Epitalon induces the activation of ribosomal genes, decondensation of pericentromeric structural heterochromatin and the release of genes repressed due to the age-related condensation of euchromatic chromosome regions. CONCLUSIONS: Epitalon has shown its ability to activate chromatin by modifying heterochromatin and heterochromatinized chromosome regions in the cells of older persons.

In the course of aging both monkeys and people reveal decreased night and average daily level of melatonin in the blood plasma and reduced hormone circadian rhythm amplitude, which evidence the disorder of the pineal gland melatonin releasing function. Peptide preparations of the pineal gland (Epithalamin--a complex of peptides isolated from the pineal gland and Epitalon--synthetic tetrapeptide) recover night release of endogenous melatonin and lead to the normalization of the hormone circadian rhythm in the blood plasma. In elderly people Epithalamin and Epitalon modulate pineal gland functional state: people with pineal gland functional insufficiency report an increase of night melatonin level. The preparations of the pineal gland, effectively increasing melatonin concentration and having no side effects, can be used in clinical geriatric practice.

The circumhoralian rhythm of protein synthesis was determined in a monolayer culture of hepatocytes from rats at the age of 1 to 24 months and weighing from 45 to 480 g, respectively. The peptide lyvagen (Lys-Glu-Asp-Ala) obtained by directed chemical synthesis on the basis of amino acid analysis of the liver polypeptide preparations increased the level of protein synthesis in the hepatocytes from rats of different ages; the highest effect was observed in the cells of old animals. In old rats, lyvagen increased the amplitude of protein synthesis fluctuations. The peptide epitalon (Ala-Glu-Asp-Gly) constructed on the basis of analysis of the epiphysis peptides did not change the intensity of protein synthesis in the cultured hepatocytes.
